Multivalent dendrons for high-affinity adhesion of proteins to DNA.

چکیده
Protein functionalization constitutes a particularly interesting target for biopharmaceutical applications, which increasingly relies on monodisperse proteinpolymer conjugates having controllable therapeutic properties.[1,2] Proteins modified with precisely controlled polymeric materials have the potential to exhibit a wide range of improved biological effects.[3] Linear polymers have been studied as part of proteinpolymer hybrids, however they induce a degree of heterogeneity in form of both the attached polymer and often the protein attachment sites, resulting in varying biochemical properties. Dendritic macromolecules offer several advantages over linear polymers, for example, a precisely defined branched structure with a high density of functional surface groups that can offer multiple simultaneous interactions leading to enhanced binding – the principle known as multivalency.[4,5]

Double-degradable responsive self-assembled multivalent arrays--temporary nanoscale recognition between dendrons and DNA.
This article reports self-assembling dendrons which bind DNA in a multivalent manner. The molecular design directly impacts on self-assembly which subsequently controls the way these multivalent nanostructures bind DNA--this can be simulated by multiscale modelling. Incorporation of an S-S linkage between the multivalent hydrophilic dendron and the hydrophobic units responsible for self-assembl...
متن کاملIn silico design a multivalent epitope vaccine against SARS-CoV-2 for Iranian populations
Background: Due to high genetic variation in human leukocyte antigen )HLA( alleles, epitope-based vaccines don’t show equal efficacy in different human populations. therefore, we proposed a multi-epitope vaccine against SARS-CoV-2 for Iranian populations. Materials and Methods: For this purpose, the proteins without allergenicity and high antigenicity as well as conservancy level from SARS-CoV...
متن کاملFacile Supermolecular Aptamer Inhibitors of L-Selectin
Multivalent interactions occur frequently in nature, where they mediate high-affinity interactions between cells, proteins, or molecules. Here, we report on a method to generate multivalent aptamers (Multi-Aptamers) that target L-selectin function using rolling circle amplification (RCA). We find that the L-selectin Multi-Aptamers have increased affinity compared to the monovalent aptamer, are ...
متن کاملHeparin versus DNA: Chiral Preferences in Polyanion Binding to Self-Assembled Multivalent (SAMul) Nanostructures.
This communication presents simple cationic self-assembling multivalent (SAMul) first generation dendrons based on L or D lysine, which form identical nanoscale assemblies in terms of dimensions and charge densities but toward which DNA and heparin exhibit different chiral binding preferences. However, higher generation dendrons with larger hydrophilic head groups are bound identically by these...
متن کاملOptimized solid phase-assisted synthesis of dendrons applicable as scaffolds for radiolabeled bioactive multivalent compounds intended for molecular imaging.
Dendritic structures, being highly homogeneous and symmetric, represent ideal scaffolds for the multimerization of bioactive molecules and thus enable the synthesis of compounds of high valency which are e.g., applicable in radiolabeled form as multivalent radiotracers for in vivo imaging. As the commonly applied solution phase synthesis of dendritic scaffolds is cumbersome and time-consuming, ...
